Thyrotropin-releasing Hormone Stimulation of Adrenocorticotropin Production

چکیده

pituitary tumors in situ, AtT-20/NYU-1 cells were found to produce the alpha subunit of the glycoprotein hormones (alpha). Alpha accumulation in the medium was constant (3.1 ng/mg cell protein per h) and was not affected by TRH. In contrast, TRH increased the amount of ACTH accumulated in the medium from AtT-20/NYU-1 cells to 190 and 420% of control at 1 and 24 h, respectively. TRH induced a dose-dependent increase in ACTH release during a 30-min incubation; half-maximal stimulation occurred at -0.1 nM. TRH had no effect on ACTH release in vitro from anterior pituitary cells derived from normal rats. Because TRH stimulates release ofACTH in some untreated patients with Cushing's disease and Nelson's syndrome as well as pathological states associated with pituitary tumors This work was presented in part at the 61st Annual Meeting of the Endocrine Society, Anaheim, Calif. 13-15 June 1979. Dr. Gershengom is the recipient of Research Career Development Award AM 00566. Received for publication 2 July 1979 and in revised form 21 January 1980. 1294 (but not in normal subjects), AtT-20/NYU-1 cells may serve as an important in vitro model for human pituitary ACTH-secreting adenomas. Moreover, these findings suggest that the primary abnormality in Cushing's disease and Nelson's syndrome, allowing TRH stimulation of ACTH release, may be intrinsic to neoplastic adrenocorticotrophs rather than in neuroregulation of ACTH release.

The Therapeutic Effect of Thyrotropin Releasing Hormone in Amyotrophic Sclerosis

Although the issue has not yet been fully resolved, there is reason to believe that TRI-I may have a trophic effect on the normal motoneuron and may transien tly improve the neurologic deficit in ALS. Since ALS is not a simple TRI-I deficiency disease, this improvement probably occurs via a secondary mecha -nism-possibly another neurotransmitter. It is uncertain at this time whether this effec...

Receptor-binding affinities and biological activities of analogs of thyrotropin-releasing hormone in prolactin-producing pituitary cells in culture.

Thyrotropin-releasing hormone (TRH) is a hypothalamic tripeptide with the structure L-pyroglutamyl-r.-histidyl-r.prolinamide. TRH stimulates prolactin and decreases growth hormone production by GH, cells, a clonal strain of rat pituitary cells grown in culture. TRH binds to specific receptors on GH, ceils. The activities of 26 analogs of TRH have been measured with the GHI cell system. Both bio...

Binding of thyrotropin-releasing hormone to plasma membranes of bovine anterior pituitary gland (hormone receptor-adenylate cyclase-equilibrium constant-( 3 H)thyrotropin).

An assay for the binding of [(3)H]thyrotropin-releasing hormone ([(3)H]TRH) is described. Plasma membranes isolated from bovine anterior pituitary gland bind about 600 femtomoles of this hormone per mg of protein, as compared to 15 femtomoles per mg of protein in the total adenohypophyseal homogenate (40-fold purification). The equilibrium constant of membrane receptor-[(3)H]TRH binding at 0 de...

Pituitary response to thyrotropin releasing hormone in children with overweight and obesity

Thyroid stimulating hormone (TSH) concentrations in the high normal range are common in children with overweight and obesity, and associated with increased cardiovascular disease risk. Prior studies aiming at unravelling the mechanisms underlying these high TSH concentrations mainly focused on factors promoting thyrotropin releasing hormone (TRH) production as a cause for high TSH concentration...

Secondary amenorrhea in a woman with spinocerebellar degeneration treated with thyrotropin-releasing hormone: a case report and in vitro analysis

INTRODUCTION While thyrotropin-releasing hormone is known to be a prolactin-release stimulating factor, thyrotropin-releasing hormone-tartrate and its derivative, taltirelin hydrate, are used for the treatment of spinocerebellar degeneration, a degenerative disease characterized mainly by motor ataxia. We report the case of a patient being treated with a thyrotropin-releasing hormone preparatio...
